Isomalt as a direct-compression excipient
Low hygroscopicity, clean flow and stability. That makes it a default in tablets and lozenges.
Excipients rarely get chosen for taste, yet for chewables, lozenges and orally disintegrating formats taste decides whether the patient finishes the course. Isomalt is one of the few bulk excipients that scores on both sides of that ledger: it compresses directly without granulation and it is pleasant to hold in the mouth. Here is why formulators keep reaching for it.
Direct compression, no granulation
Fine-grade isomalt (NT-ST-80 and NT-ST-150) has a particle size distribution and surface texture that let it flow freely through hoppers and compress at 5 to 15 kN into hard, low-friability tablets. Skipping wet granulation removes a drying step, a solvent and a source of batch variation. For moisture-sensitive actives it also removes the water entirely.
Very low hygroscopicity
Isomalt takes up little moisture under normal storage conditions. In a blister or bottle this means the tablet does not soften, the active does not hydrolyse and the shelf life claim survives an Indian summer. Compare sorbitol, which starts taking on water at around 70% RH.
For a chewable or lozenge, the excipient is the dosage form. Patients taste it before they feel the active.
Chemically inert
As a hydrogenated sugar alcohol, isomalt has no reducing groups. It will not undergo Maillard reactions with amine-containing actives, so no browning and no adduct formation on stability. It is stable across pH 2 to 10 and through the temperatures seen in coating pans and lozenge kettles.
The mouth experience
Isomalt is about half as sweet as sucrose with a mild cooling effect and no aftertaste. For chewables and lozenges that is a clean canvas: sweetness is topped up with a high-intensity sweetener and flavours read true. It is also non-cariogenic, which matters for products held in the mouth for minutes at a time.
Where to start
Blend NT-ST-80 with the active and 0.5 to 1% magnesium stearate, compress and test. For lozenges, cook isomalt to 160 to 165 °C, add the active below 120 °C and deposit. Batch COAs and full specifications ship with every sample.
Key takeaways
- 01NT-ST-80 compresses directly at 5 to 15 kN; no granulation step.
- 02Low moisture uptake helps protect sensitive actives.
- 03No reducing groups, so no Maillard reactions with amine actives.
- 04Mild, cooling, non-cariogenic sweetness for formats held in the mouth.
